The interpretation of the law of definite proportions in terms of the atomic theory of matter implied that the masses of atoms of various elements had definite ratios that depended on the elements. While the actual masses were unknown, the relative masses could be deduced from that law. In 1803 John Dalton proposed to use the (still unknown) atomic mass of the lightest atom, hydrogen, as the natural unit of atomic mass. This was the basis of the atomic weight scale. For technical reasons, in 1898, chemist Wilhelm Ostwald and others proposed to redefine the unit of atomic mass as 1/16 the mass of an oxygen atom. That proposal was formally adopted by the International Committee on Atomic Weights (ICAW) in 1903. That was approximately the mass of one hydrogen atom, but oxygen was more amenable to experimental determination. This suggestion was made before the discovery of isotopes in 1912. === Significance === In some experiments, the charge-to-mass ratio is the only quantity that can be measured directly. Often, the charge can be inferred from theoretical considerations, so the charge-to-mass ratio provides a way to calculate the mass of a particle. Often, the charge-to-mass ratio can be determined by observing the deflection of a charged particle in an external magnetic field. The cyclotron equation, combined with other information such as the kinetic energy of the particle, will give the charge-to-mass ratio. One application of this principle is the mass spectrometer. The same principle can be used to extract information in experiments involving the cloud chamber. The ratio of electrostatic to gravitational forces between two particles will be proportional to the product of their charge-to-mass ratios. It turns out that gravitational forces are negligible on the subatomic level, due to the extremely small masses of subatomic particles.
In laboratories and industry, vacuum flasks are often used to hold liquefied gases (commonly liquid nitrogen with a boiling point of 77 K) for flash freezing, sample preparation and other processes where creating or maintaining an extreme low temperature is desired. Larger vacuum flasks store liquids that become gaseous at well below ambient temperature, such as oxygen and nitrogen; in this case the leakage of heat into the extremely cold interior of the bottle results in a slow boiling-off of the liquid so that a narrow unstoppered opening, or a stoppered opening protected by a pressure relief valve, is necessary to prevent pressure from building up and eventually shattering the flask. The insulation of the vacuum flask results in a very slow "boil" and thus the contents remain liquid for long periods without refrigeration equipment. Vacuum flasks have been used to house standard cells and ovenized Zener diodes, along with their printed circuit board, in precision voltage-regulating devices used as electrical standards. The flask helped with controlling the Zener temperature over a long time span and was used to reduce variations of the output voltage of the Zener standard owing to temperature fluctuation to within a few parts per million. === Positive selection === T cells have distinct receptors. These receptors are formed by the process of V(D)J recombination gene rearrangement stimulated by RAG1 and RAG2 genes. This process is error-prone, and some thymocytes do not make functional T-cell receptors, whereas other thymocytes make autoreactive T-cell receptors. If a functional T cell receptor is formed, the thymocyte expresses the cell surface proteins CD4 and CD8 simultaneously. The survival and nature of the T cell then depend on its interaction with surrounding thymic epithelial cells. The T cell receptor interacts with the MHC molecules on the surface of epithelial cells. A T cell with a receptor that doesn't react, or reacts weakly, dies by apoptosis. T cells that react survive and proliferate. A mature T cell expresses either CD4 or CD8, but not both. This depends on the binding strength between the TCR and MHC class 1 or class 2. A T cell receptor that binds mostly to MHC class I tends to produce a mature "cytotoxic" CD8 positive T cells those that bind mostly to MHC class II typically produce a CD4 positive T cell.
