Semaglutide is a synthetic peptide of thirty-one amino acids that shares roughly ninety-four percent sequence identity with human glucagon-like peptide-1. Two substitutions resist enzymatic cleavage by dipeptidyl peptidase-4, and a fatty diacid side chain attached through a linker promotes binding to serum albumin. That albumin binding slows renal clearance and extends the circulating half-life from minutes to approximately one week. The structural changes are well established in the published literature. Whether the same modifications affect receptor signalling bias in ways that matter clinically remains an open question.
Pharmacological activity arises from agonism at the glucagon-like peptide-1 receptor, a G protein-coupled receptor expressed in the pancreas, the gastrointestinal tract, and the brainstem. Receptor activation raises intracellular cyclic adenosine monophosphate and enhances insulin release in a glucose-dependent manner, an effect that diminishes when blood glucose concentration is low. Other effects include slowed gastric emptying and hypothalamic satiety signalling. These pathways are described well. Receptor desensitisation rates across tissues, relative to the endogenous hormone, are still under investigation, and reported findings differ between laboratories.
The company that developed the compound filed it as a long-acting analogue, and it gained first approval in 2017 for type 2 diabetes. Later authorisations from several regulators extended the indication to chronic weight management, and the World Health Organization added the glucagon-like peptide-1 receptor agonist drug class to its model list of essential medicines in 2023. Production uses solid-phase peptide synthesis followed by side-chain conjugation and chromatographic purification. Supply constraints and cost differences across regions are well documented. Literature on long-term outcomes continues to grow, with many trials reporting surrogate endpoints rather than hard clinical endpoints.
Peptides are sensitive to temperature, light, oxygen, and repeated freeze-thaw cycles. Semaglutide in dry form is generally held at refrigerated temperatures, while reconstituted solutions require a defined short-term storage window. Vials should be kept in secondary packaging to limit photodegradation, and exposure to alkaline conditions is avoided because it accelerates chemical degradation. Adsorption to glass and some plastics can reduce the measured concentration of dilute solutions, so low-binding polypropylene containers are preferred for analytical work. Each transfer step introduces a small risk of contamination, and closed handling practices reduce that risk.
Routine characterisation of the peptide relies on reversed-phase high-performance liquid chromatography, often paired with ultraviolet detection near 214 nanometres. Related substances such as deamidated, oxidised, and truncated sequences elute at characteristic positions and are quantified by area percentage. Electrospray ionisation mass spectrometry confirms the molecular mass and can resolve some closely related variants. Peptide mapping after enzymatic digestion provides sequence-level verification and is useful when a full identity profile is required. Method parameters such as column chemistry, gradient, and mobile-phase pH influence the separation and must be reported alongside results.
Material described as research-grade is not necessarily manufactured to pharmaceutical standards, and purity figures depend on the method used to obtain them. A certificate of analysis states the measured purity, the analytical technique, and the batch identifier, but the underlying data are not always included. Independent testing by a second laboratory is a common way to confirm identity and purity. Uncertainties remain about how storage history affects long-term stability, and about how well results from one laboratory transfer to another. Documentation of handling conditions supports comparison between batches.
| Property | Value | Notes |
|---|---|---|
| Molecular class | 31-amino-acid peptide | Modified glucagon-like peptide-1 analogue |
| Molecular mass | approximately 4114 Da | Calculated for the free peptide backbone |
| Appearance | white to off-white powder | Typical of purified lyophilised peptide batches |
| Solubility class | freely soluble in water | Aqueous solubility improves at slightly alkaline pH |
| Typical storage | -20 degrees Celsius, dry, dark | Protect from repeated freeze-thaw cycles |
Storage at minus 20 degrees Celsius or lower in a desiccated container preserves the peptide for extended periods, while working solutions are commonly held at two to eight degrees Celsius for short intervals. Light exposure and repeated freeze-thaw cycles accelerate degradation, so dividing material into single-use aliquots is generally recommended. Adsorption to glass and plastic surfaces can lower the measured concentration of dilute solutions, particularly below one milligram per millilitre. The degradation routes most often reported for GLP-1 analogues are deamidation, methionine oxidation, and backbone hydrolysis. Relative rates under specific conditions are frequently described only for individual formulations.
Reverse-phase high-performance liquid chromatography with ultraviolet detection near 214 or 280 nanometres is widely used to assess purity and to resolve related impurities. Liquid chromatography coupled to mass spectrometry confirms identity through the protonated molecular ion and fragment ions formed in tandem experiments. Capillary electrophoresis and peptide mapping after enzymatic digestion supply complementary information on charge variants and modification sites. Circular dichroism and nuclear magnetic resonance can report on secondary structure in solution. Absolute quantification usually depends on an external standard, and reported purity depends on the detection wavelength and integration parameters chosen.
Native GLP-1 is degraded rapidly by dipeptidyl peptidase-4. Semaglutide resists this cleavage because alanine at position 8 is replaced by alpha-aminoisobutyric acid. A second substitution at position 34 introduces arginine, which further stabilizes the peptide. The most distinctive modification is a spacer and C18 fatty diacid attached at lysine 26, enabling strong albumin affinity. These three changes together produce a half-life measured in days rather than minutes, and the same structural logic underlies other long-acting analogs in this class.
Semaglutide is a synthetic peptide analog of glucagon-like peptide-1, a hormone released from intestinal L-cells after food intake. It contains 31 amino acids and differs from native GLP-1 through modifications that slow enzymatic breakdown. The peptide was developed to extend the short circulating half-life of endogenous GLP-1, which is measured in minutes. Researchers introduced the compound in the early 2010s. Two backbone changes and a fatty acid side chain define its structure, distinguishing it from earlier GLP-1 receptor agonists.
The compound binds the GLP-1 receptor on pancreatic beta cells and other tissues, activating a G-protein signaling cascade that raises intracellular cyclic AMP. This action increases glucose-dependent insulin secretion when blood glucose is elevated, while binding also slows gastric emptying and reduces glucagon release. In the central nervous system, receptor activation in the hypothalamus and brainstem contributes to reduced appetite. The fatty acid chain binds albumin, which protects the peptide from renal filtration and enzymatic degradation. This albumin binding is central to its extended circulation time.
==== Immunotherapy ==== Dysfunction of the immune system can cause autoimmune diseases, inflammatory diseases and cancer. Immunodeficiency occurs when the immune system is less active than normal, resulting in recurring and life-threatening infections, and can be the result of a genetic disease such as severe combined immunodeficiency, acquired conditions such as HIV/AIDS, or the use of immunosuppressive medication. Autoimmunity describes a hyperactive immune system attacking normal tissues as if they were foreign organisms; diseases include Hashimoto's thyroiditis, rheumatoid arthritis, diabetes mellitus type 1, and systemic lupus erythematosus.
MDMA is being investigated as a potential treatment for social impairments in autism. The British critical psychiatrist Joanna Moncrieff has critiqued the use and study of MDMA and related drugs like psychedelics for treatment of psychiatric disorders, highlighting concerns including excessive hype around these drugs, blurred lines between medical and recreational use, flawed clinical trial findings, financial conflicts of interest, strong expectancy effects and large placebo responses, short-term benefits over placebo, and their potential for adverse effects, among others.

=== Distributor's losses and impact === Though Darbar became the highest-grossing Tamil film of 2020, grossing ₹202–250, several distributors claimed that they incurred significant financial losses due to the film’s high acquisition costs and pre-release rights pricing. Trade analysts estimated that distributors collectively suffered losses of around ₹70 crore. It did not perform well in other Indian states, including Andhra Pradesh and Telangana, due to competition from Ala Vaikunthapurramuloo and Sarileru Neekevvaru, and the Hindi version was unsuccessful at the box office due to a lack of promotion. Following unsuccessful discussions with Lyca Productions, a group of distributors attempted to approach Rajinikanth seeking financial relief. Media reports stated that some distributors threatened protests, including hunger strikes, if their concerns were not addressed. Industry commentators later compared the situation to earlier Rajinikanth films such as Baba (2002), Kuselan (2008) and Lingaa (2014). Trade publications attributed the dispute primarily to the film’s high production and acquisition costs. Reports indicated that Rajinikanth and director A. R. Murugadoss received substantial remuneration, contributing to an increased production budget, while the film’s distribution rights were sold at premium rates. Some commentators cited the episode as part of a broader industry debate on the financial risks associated with high-budget star-driven films.
In the case of feminizing surgery surgery for transgender patients, labiaplasty is usually the second stage of a two-stage vaginoplasty/vulvoplasty operation, where labiaplasty techniques are applied to create labia minora and a clitoral hood. In this procedure, the labiaplasty is usually performed some months after the first stage of vaginoplasty.
Adrenodoxin-NADP+ reductase (EC 1.18.1.6, adrenodoxin reductase, nicotinamide adenine dinucleotide phosphate-adrenodoxin reductase, ADR, NADPH:adrenal ferredoxin oxidoreductase) is an enzyme with systematic name adrendoxin:NADP+ oxidoreductase. This enzyme catalyses the following chemical reaction
=== Chemical lysis === This method uses chemical disruption. It is the most popular and simple approach. Chemical lysis chemically deteriorates/solubilizes the proteins and lipids present within the membrane of targeted cells. Common lysis buffers contain sodium hydroxide (NaOH) and sodium dodecyl sulfate (SDS). Cell lysis is best done at a pH range of 11.5–12.5. Although simple, it is a slow process, taking anywhere from 6 to 12 hours.

Hours after the capture, the United States attorney general Pam Bondi announced that Maduro and Flores had been indicted in the Southern District of New York on charges related to "narcoterrorism". The indictment listed the charges as narco-terrorism conspiracy, cocaine importation conspiracy, possession of machine guns and destructive devices, and conspiracy to possess machine guns and destructive devices. The amended indictment did not refer to the Cartel of the Suns as an organized group as previously alleged by the US, but instead as a corrupt patronage system, which aligns with experts' descriptions of its operations. Maduro and his wife were arraigned in a Manhattan federal court on 5 January 2026. Before pleading, Maduro said: "I am the president of Venezuela, I consider myself a prisoner of war. I was captured at my home in Caracas." Both pleaded not guilty to numerous drug trafficking charges. US District Judge Alvin Hellerstein ordered that Maduro be held at least until a hearing. Originally scheduled for 17 March, the hearing was delayed to 26 March. Maduro and Flores appeared in court for it, then were transferred back to jail in Brooklyn. Maduro is reportedly spending most of his time in prison reading the Bible. He does not have access to newspapers or the internet in prison, but he is allowed to speak by telephone with his family and lawyers for approximately 300 minutes a month, with a maximum of 15 minutes per call. Their trial was scheduled for 1 June 2027.
==== Fluorescent (nitrogen vacancy) ==== Fluorescent properties in nanodiamonds arise from the presence of nitrogen-vacancy (NV) centers, nitrogen atoms next to a vacancy. Fluorescent nanodiamond (FND) was invented in 2005 and has since been used in various fields of study. The invention received a US patent in 2008 States7326837 B2 United States 7326837 B2, Chau-Chung Han; Huan-Cheng Chang & Shen-Chung Lee et al., "Clinical applications of crystalline diamond particles", issued February 5, 2008, assigned to Academia Sinica, Taipei (TW) , and a subsequent patent in 2012 States8168413 B2 United States 8168413 B2, Huan-Cheng Chang; Wunshian Fann & Chau-Chung Han, "Luminescent Diamond Particles", issued May 1, 2012, assigned to Academia Sinica, Taipei (TW) . NV centers can be created by irradiating nanodiamonds with high-energy particles (electrons, protons, helium ions), followed by vacuum-annealing at 600–800 °C. Irradiation forms vacancies in the diamond structure while vacuum-annealing migrates these vacancies, which will get trapped by nitrogen atoms within the nanodiamond. This process produces two types of NV centers. Two types of NV centers are formed—neutral (NV0) and negatively charged (NV–)—and these have different emission spectra. The NV– the center is of particular interest because it has an S = 1 spin ground state that can be spin-polarized by optical pumping and manipulated using electron paramagnetic resonance.
In 2000, the chapter on Vitamin C in the North American Dietary Reference Intake was updated to give the Recommended Dietary Allowance (RDA) as 90 milligrams per day for adult men, 75 mg/day for adult women, and setting a tolerable upper intake level (UL) for adults of 2,000 mg/day. The table here shows RDAs for the United States and Canada for children, and for pregnant and lactating women, as well as the ULs for adults. For the European Union, the EFSA set higher recommendations for adults, and also for children: 20 mg/day for ages 1–3, 30 mg/day for ages 4–6, 45 mg/day for ages 7–10, 70 mg/day for ages 11–14, 100 mg/day for males ages 15–17, 90 mg/day for females ages 15–17. For pregnancy 100 mg/day; for lactation 155 mg/day. Cigarette smokers and people exposed to secondhand smoke have lower serum vitamin C levels than nonsmokers. The reasoning is that inhalation of smoke causes oxidative damage, depleting this antioxidant vitamin. The US Institute of Medicine estimated that smokers need 35 mg more vitamin C per day than nonsmokers, but did not formally establish a higher RDA for smokers. The US National Center for Health Statistics conducts biannual National Health and Nutrition Examination Survey (NHANES) to assess the health and nutritional status of adults and children in the United States. Some results are reported as What We Eat In America. The 2013–2014 survey reported that for adults ages 20 years and older, men consumed on average 83.3 mg/d and women 75.1 mg/d. This means that half the women and more than half the men are not consuming the RDA for vitamin C.
=== Neovascular glaucoma === Anti-VEGF agents as injectable medications, along with other standard of care treatment for decreasing intraocular pressure, may improve pressure in people with neovascular glaucoma for short periods. Evidence suggests that this improvement may last 4–6 weeks. There is no sufficient evidence to suggest that anti-VEGF medications are effective either for short-term or for longer-term treatment. The short, medium, and long-term safety of anti-VEGF treatment has not been well investigated.

It is a glucagon-like peptide-1 receptor agonist, often grouped with the incretin mimetics. Its backbone is modified from the human hormone to resist enzymatic degradation and to bind albumin. These two features distinguish it from the native peptide.
No. It shares high sequence identity but carries non-natural modifications that alter its stability and clearance. The naturally occurring hormone is broken down within minutes, while the synthetic analogue circulates far longer.
Attachment to serum albumin reduces renal filtration of the peptide and shields it from enzymatic breakdown. The consequence is a much longer interval between administrations than unmodified peptides allow. The precise contribution of binding affinity to overall duration is still being quantified.
Different techniques detect different classes of impurities, so a single number does not describe a sample completely. Reversed-phase chromatography resolves related peptides well but can miss inorganic salts, while mass spectrometry confirms mass without quantifying everything present. Comparing results requires knowing which method was used and how it was validated.
