Native GLP-1 is degraded rapidly by dipeptidyl peptidase-4. Semaglutide resists this cleavage because alanine at position 8 is replaced by alpha-aminoisobutyric acid. A second substitution at position 34 introduces arginine, which further stabilizes the peptide. The most distinctive modification is a spacer and C18 fatty diacid attached at lysine 26, enabling strong albumin affinity. These three changes together produce a half-life measured in days rather than minutes, and the same structural logic underlies other long-acting analogs in this class.
Semaglutide is a synthetic peptide analog of glucagon-like peptide-1, a hormone released from intestinal L-cells after food intake. It contains 31 amino acids and differs from native GLP-1 through modifications that slow enzymatic breakdown. The peptide was developed to extend the short circulating half-life of endogenous GLP-1, which is measured in minutes. Researchers introduced the compound in the early 2010s. Two backbone changes and a fatty acid side chain define its structure, distinguishing it from earlier GLP-1 receptor agonists.
The compound binds the GLP-1 receptor on pancreatic beta cells and other tissues, activating a G-protein signaling cascade that raises intracellular cyclic AMP. This action increases glucose-dependent insulin secretion when blood glucose is elevated, while binding also slows gastric emptying and reduces glucagon release. In the central nervous system, receptor activation in the hypothalamus and brainstem contributes to reduced appetite. The fatty acid chain binds albumin, which protects the peptide from renal filtration and enzymatic degradation. This albumin binding is central to its extended circulation time.
Lyophilised semaglutide is generally held at -20 °C or below, protected from light and moisture. Reconstituted solutions are typically kept at 2-8 °C and used within a defined window because degradation accumulates over time. Repeated freeze-thaw cycles are discouraged, since each cycle can promote aggregation and reduce monomeric content. Room-temperature stability of the solid has been examined in some studies but remains incompletely characterised for long durations, so cold storage is the conservative default for research material.
Degradation proceeds along several parallel routes. Deamidation of asparagine and glutamine residues generates charged variants that shift retention time in chromatographic analysis. Oxidation targets methionine and can be accelerated by trace metals or dissolved oxygen. Non-covalent aggregation produces dimers, oligomers, and larger species that are difficult to reverse. Isomerisation at aspartate residues is slower but measurable under thermal stress. The distribution among these pathways depends on pH, buffer composition, ionic strength, and the presence of excipients such as sugars or surfactants.

Routine characterisation of the peptide relies on reversed-phase high-performance liquid chromatography, often paired with ultraviolet detection near 214 nanometres. Related substances such as deamidated, oxidised, and truncated sequences elute at characteristic positions and are quantified by area percentage. Electrospray ionisation mass spectrometry confirms the molecular mass and can resolve some closely related variants. Peptide mapping after enzymatic digestion provides sequence-level verification and is useful when a full identity profile is required. Method parameters such as column chemistry, gradient, and mobile-phase pH influence the separation and must be reported alongside results.
Material described as research-grade is not necessarily manufactured to pharmaceutical standards, and purity figures depend on the method used to obtain them. A certificate of analysis states the measured purity, the analytical technique, and the batch identifier, but the underlying data are not always included. Independent testing by a second laboratory is a common way to confirm identity and purity. Uncertainties remain about how storage history affects long-term stability, and about how well results from one laboratory transfer to another. Documentation of handling conditions supports comparison between batches.
Peptides are sensitive to temperature, light, oxygen, and repeated freeze-thaw cycles. Semaglutide in dry form is generally held at refrigerated temperatures, while reconstituted solutions require a defined short-term storage window. Vials should be kept in secondary packaging to limit photodegradation, and exposure to alkaline conditions is avoided because it accelerates chemical degradation. Adsorption to glass and some plastics can reduce the measured concentration of dilute solutions, so low-binding polypropylene containers are preferred for analytical work. Each transfer step introduces a small risk of contamination, and closed handling practices reduce that risk.
Clinical studies of semaglutide generally measure glycated hemoglobin, fasting plasma glucose, body weight, and composite cardiovascular endpoints. The SUSTAIN program enrolled adults with type 2 diabetes, while the STEP program focused on obesity without diabetes. Administration follows a stepwise escalation schedule designed to limit gastrointestinal effects during the first weeks. Reported outcomes include mean percentage weight change, the proportion of participants reaching defined weight-loss thresholds, and rates of nausea, vomiting, and diarrhea. Long-term data on durability after treatment stops are still limited and remain a topic of ongoing research.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, developed by Novo Nordisk and first approved in 2017 for type 2 diabetes. It belongs to the incretin mimetic class, a group of agents that reproduce the glucose-dependent actions of endogenous GLP-1. The molecule was engineered to resist degradation by dipeptidyl peptidase-4 and to bind serum albumin, extending its half-life from minutes to roughly one week. Approval for chronic weight management followed in 2021, based on large cardiovascular and obesity outcome trials.

=== Space science and planetary research === Space science and planetary research at the University of Arizona are largely conducted through the Lunar and Planetary Laboratory (LPL), which has led or contributed to numerous NASA missions. The university received more than $325 million for the LPL to lead NASA's 2007–08 Phoenix Mars Mission, which explored the Martian Arctic. It later received another $800 million for the OSIRIS-REx mission, the first mission in U.S. history to collect a sample from an asteroid. The LPL's contributions to the Cassini mission orbiting Saturn were larger than those of any other university in the world. University researchers designed and operated the spacecraft's atmospheric radiation and imaging investigations. The university also operates the HiRISE camera aboard the Mars Reconnaissance Orbiter. The spacecraft reached Mars in March 2006 after traveling 300 million miles. The University of Arizona-designed camera was led by planetary scientist Alfred McEwen, the project's principal investigator. HiRISE captured high-resolution images of the planet. In 2011, University of Arizona alumnus and planetary scientist Lujendra Ojha and his research team used HiRISE imagery to identify evidence of liquid water on the surface of Mars. NASA confirmed the discovery in 2015. The university receives more NASA grants annually than the next nine top NASA-Jet Propulsion Laboratory-funded universities combined. In August 2007, the University of Arizona, under the direction of planetary scientist Peter Smith, led the Phoenix Mars Mission.
Marketers use well-researched techniques to try to control purchasing behavior. The layout of a supermarket is considered by some to consist of a few rules of thumb and three layout principles. The high-draw products are placed in separate areas of the store to keep drawing the consumer through the store. High impulse and high margin products are placed in the most predominant areas to grab attention. Power products are placed on both sides of the aisle to create increased product awareness, and end caps are used to receive a high exposure of a certain product whether on special, promotion or in a campaign, or a new line. The first principle of the layout is circulation. Circulation is created by arranging product so the supermarket can control the traffic flow of the consumer. Along with this path, there will be high-draw, high-impulse items that will influence the consumer to make purchases which they did not originally intend. Service areas such as restrooms are placed in a location which draws the consumer past certain products to create extra buys. Necessity items such as bread and milk are found at the rear of the store to increase the start of circulation. Cashiers' desks are placed in a position to promote circulation. In most supermarkets, the entrance will be on the right-hand side because some research suggests that consumers who travel in a counter-clockwise direction spend more. However, other researchers have argued that consumers moving in a clockwise direction can form better mental maps of the store leading to higher sales in turn.

=== Improved diagnosis and treatment === The first attempt to establish a set of diagnostic criteria was also due to Charcot in 1868. He published what now is known as the "Charcot triad", consisting of nystagmus, intention tremor, and telegraphic speech (scanning speech). Charcot also observed cognition changes, describing his patients as having a "marked enfeeblement of the memory" and "conceptions that formed slowly". The diagnosis was based on Charcot triad and clinical observation until Schumacher made the first attempt to standardize criteria in 1965 by introducing some fundamental requirements: Dissemination of the lesions in time (DIT) and space (DIS), and that "signs and symptoms cannot be explained better by another disease process". The DIT and DIS requirement was later inherited by the Poser and McDonald criteria. During the 20th century, theories about the cause and pathogenesis were developed and effective treatments began to appear in the 1990s. Since the beginning of the 21st century, refinements of the concepts have taken place. The 2010 revision of the McDonald criteria allowed for the diagnosis of MS with only one proved lesion (CIS). In 1996, the US National Multiple Sclerosis Society (NMSS) (Advisory Committee on Clinical Trials) defined the first version of the clinical phenotypes that is in use. In this first version, they provided standardized definitions for four MS clinical courses: relapsing-remitting (RR), secondary progressive (SP), primary progressive (PP), and progressive relapsing (PR). In 2010, PR was dropped and CIS was incorporated.

Living organisms must obey the laws of thermodynamics, which describe the transfer of heat and work. The second law of thermodynamics states that in any isolated system, the amount of entropy (disorder) cannot decrease. Although living organisms' amazing complexity appears to contradict this law, life is possible as all organisms are open systems that exchange matter and energy with their surroundings. Living systems are not in equilibrium, but instead are dissipative systems that maintain their state of high complexity by causing a larger increase in the entropy of their environments. The metabolism of a cell achieves this by coupling the spontaneous processes of catabolism to the non-spontaneous processes of anabolism. In thermodynamic terms, metabolism maintains order by creating disorder.
Acral angiofibromas are also termed superficial acral fibromyxomas, digital fibromyxomas, acquired digital fibrokeratomas, acquired periungual fibrokeratomas, garlic clove fibromas, digital fibromas, and cellular digital fibromas. At one time, periungual angiofibromas were regarded as a type of acral angiofibroma (see above description). Acral refers to distal sites of the ears, nose, hands, fingers, feet, and toes. Acral angifibromeae occur primarily in areas close to the nails of fingers and toes (~80% of cases) or, less commonly, palms of the hands or soles of the feet. The tissues of this tumor consists of bland spindle-shaped and star-shaped cells within a collagen fiber-rich stroma containing prominent blood vessels and mast cells.
== Mechanism == The main function of the SEC23A protein is to hydrolyze or break down a guanosine triphosphate (GTP) molecule bound to the SAR1A protein at the start of the COPII pathway. The energy released from the breaking of the GTP bond provides energy necessary to undergo another reaction. This triggers uncoating of the vesicle (a membrane bound carrying compartment for molecules) containing a secretory protein destined for packaging in the Golgi apparatus of the cell. Uncoating the vesicle exposes SNARE proteins which are needed for the vesicle to bind to the membrane site on the endoplasmic reticulum. A mutation in the SEC23A gene prevents the vesicle from uncoating so it will not bind to the receptor site on the endoplasmic reticulum to be released into the cytoplasm for transport to the Golgi apparatus. Thus, the vesicles will accumulate in the endoplasmic reticulum, causing it to become enlarged or distended. Ultimately, this causes the craniofacial symptoms present in patients with CLSD. This is probably due to abnormal secretion of collagen and possibly other secretory proteins which have accumulated in the endoplasmic reticulum. Collagen is responsible to for skull ossification, among other things.
==== Hydrides and halides ==== The hydride InH3 has at best a transitory existence in ethereal solutions at low temperatures. It polymerizes in the absence of bases. Lewis bases stabilize a rich collection of indium hydrides of the formula LInH3 (L = tertiary phosphine and N-Heterocyclic carbenes). Chlorination, bromination, and iodination of In produce colorless InCl3, InBr3, and yellow InI3. The compounds are Lewis acids, somewhat akin to the better known aluminium trihalides. Again like the related aluminium compound, InF3 is polymeric. Indium halides dissolves in water to give aquo complexes such as [In(H2O)6]3+ and [InCl2(H2O)4]+. Similar complexes can be prepared from nitrates and acetates. Overall, the pattern is similar to that for aluminium(III).

It is a synthetic analog of GLP-1 produced through medicinal chemistry to resist enzymatic degradation. The design goal was longer circulation than the native hormone.
A fatty acid side chain attaches the peptide to serum albumin, which shields it from kidney filtration and protease activity. This interaction is the main reason its circulation time is extended.
No. The native hormone is GLP-1, and semaglutide is an engineered variant with three deliberate structural alterations. It does not appear in unmodified biological sources.
Reverse-phase high-performance liquid chromatography with ultraviolet detection is the usual choice, with results reported as area percent. Complementary methods such as size-exclusion chromatography and mass spectrometry are needed because a single separation cannot resolve every impurity class. Purity figures are therefore method dependent and should always be read alongside the technique used.
