The study of proteins in vivo is often concerned with the synthesis and localization of the protein within the cell. Although many intracellular proteins are synthesized in the cytoplasm and membrane-bound or secreted proteins in the endoplasmic reticulum, the specifics of how proteins are targeted to specific organelles or cellular structures is often unclear. A useful technique for assessing cellular localization uses genetic engineering to express in a cell a fusion protein or chimera consisting of the natural protein of interest linked to a "reporter" such as green fluorescent protein (GFP). The fused protein's position within the cell can then be cleanly and efficiently visualized using microscopy. Other methods for elucidating the cellular location of proteins requires the use of known compartmental markers for regions such as the ER, the Golgi, lysosomes or vacuoles, mitochondria, chloroplasts, plasma membrane, etc. With the use of fluorescently tagged versions of these markers or of antibodies to known markers, it becomes much simpler to identify the localization of a protein of interest. For example, indirect immunofluorescence will allow for fluorescence colocalization and demonstration of location. Fluorescent dyes are used to label cellular compartments for a similar purpose. Other possibilities exist, as well. For example, immunohistochemistry usually uses an antibody to one or more proteins of interest that are conjugated to enzymes yielding either luminescent or chromogenic signals that can be compared between samples, allowing for localization information.

On 4 June, Anvisa approved exceptional imports of Sputnik V, restricting it mainly to healthy adults and limiting it to only 1% of the population of 6 importing states, in order to manage risks through control and supervision of side effects. Anvisa said that the concern with replicating viruses has not been fully resolved, but that additional documents received indicate a substantially reduced acceptable amount. The new parameter would be in an FDA manual, which was not found. Anvisa also said that impurity and quality controls are insufficient and that the manufacturing plants must undergo corrections to meet WHO quality standards. As of 16 June, the same import conditions were extended to a total of 13 states. On 5 August, the consortium of northeastern Brazilian states, corresponding to 7 of the 13 states, suspended the import of 37 million doses due to the restrictions imposed by Anvisa. These doses will supply Mexico, Argentina and Bolivia.
== History == The first definition of the term bioinformatics was coined by Paulien Hogeweg and Ben Hesper in 1970, to refer to the study of information processes in biotic systems. This definition placed bioinformatics as a field parallel to biochemistry (the study of chemical processes in biological systems). Bioinformatics and computational biology involved the analysis of biological data, particularly DNA, RNA, and protein sequences. The field of bioinformatics experienced explosive growth starting in the mid-1990s, driven largely by the Human Genome Project and by rapid advances in DNA sequencing technology. Analyzing biological data to produce meaningful information involves writing and running software programs that use algorithms from graph theory, artificial intelligence, soft computing, data mining, image processing, and computer simulation. ==== Musée d'histoire naturelle (Museum of Natural History) ==== Founded in 1881 but heavily damaged during World War II, the Museum of Natural History is housed in Le Havre's former law courts, built in the mid-18th century; the façade and monumental staircase are listed as historical monuments. The museum houses mineralogy, zoology, ornithology, palaeontology and prehistory departments as well as 8,000 early 19th-century paintings from the collection of local naturalist and traveller Charles-Alexandre Lesueur (1778–1846). The museum was destroyed during Allied bombings on 5 September 1944. The library was lost, along with its collections of photographs, scientific instruments and archives. The mineral and geological collections were all destroyed, including a rare collection of local mineral specimens of Normandy. The destruction of the museum was so intense, that all the catalogues, lists of donations, lists of purchases and other archives prevented even a precise inventory of all that was lost."
=== High-speed supernatant (HSS) === High-speed supernatant (HSS) is a fraction obtained by ultracentrifuging a conventional Xenopus egg extract at 100,000–200,000 × g, which removes membrane components and ribosomes, leaving a solution enriched in soluble proteins. Although HSS lacks the capacity to support nuclear assembly or protein translation, it can partially recapitulate chromatin structural changes in a cell cycle–dependent manner. It is particularly suitable for protein purification.

=== Formation of the first hypercycles === Eigen made several assumptions about conditions that led to the formation of the first hypercycles. Some of them were the consequence of the lack of knowledge about ribozymes, which were discovered a few years after the introduction of the hypercycle concept and negated Eigen's assumptions in the strict sense. The primary of them was that the formation of hypercycles had required the availability of both types of chains: nucleic acids forming a quasispecies population and proteins with enzymatic functions. Nowadays, taking into account the knowledge about ribozymes, it may be possible that a hypercycle's members were selected from the quasispecies population and the enzymatic function was performed by RNA. According to the hypercycle theory, the first primitive polymerase emerged precisely from this population. As a consequence, the catalysed replication could exceed the uncatalysed reactions, and the system could grow faster. However, this rapid growth was a threat to the emerging system, as the whole system could lose control over the relative amount of the RNAs with enzymatic function. The system required more reliable control of its constituents—for example, by incorporating the coupling of essential RNAs into a positive feedback loop. Without this feedback loop, the replicating system would be lost. These positive feedback loops formed the first hypercycles.
==== Ion exchange chromatography ==== Ion exchange chromatography is often employed in the first step, or capturing step, for the separation of PEGylated proteins as PEGylation may affect the charges of target proteins by neutralizing electrostatic interaction, changing the isoelectric point (pI), and increasing the pKa value. Due to the high pI of lysozyme (pI = 10.7), cation exchange chromatography is used. As the increased degree of PEGylation decreases the ion strength of the protein, the poly-PEGylated proteins tend to bind to the cation resin weaker than the mono-PEGylated protein or the intact form does. Thus, the poly-PEGylated proteins elute faster and the intact protein eludes last in the cation exchange chromatography. As mono-PEGylated is widely investigated and described as a protection of target proteins, the target eluate in the cation exchange chromatography is usually the mono-PEGylated proteins.
In late 19th-century Venezuela, a German-born doctor named Gottfried Knoche conducted experiments in mummification at his laboratory in the forest near La Guaira. He developed an embalming fluid (based on an aluminium chloride compound) that mummified corpses without having to remove the internal organs. The formula for his fluid was never revealed and has not been discovered. Most of the several dozen mummies created with the fluid (including himself and his immediate family) have been lost or were severely damaged by vandals and looters.
