Large randomised trials in adults with type 2 diabetes and in adults with obesity have reported reductions in body weight and improvements in several cardiovascular risk markers. One outcome trial found a lower incidence of major adverse cardiovascular events in participants with diabetes and established cardiovascular disease. Gastrointestinal effects such as nausea and vomiting are the most frequently reported adverse events and often diminish over time. Changes in lean body mass during weight loss are an area of ongoing investigation. Effects in adolescents and in pregnancy are less well characterised, and current labelling advises against use during pregnancy.
Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released after nutrient intake. Performance-enhancing substances (PESs), also known as performance-enhancing drugs (PEDs), are substances that are used to improve any form of activity performance in humans. Many substances, such as anabolic steroids, can be used to improve athletic performance and build muscle, which in most cases is considered cheating by organized athletic organizations. This usage is often referred to as doping. Athletic performance-enhancing substances are sometimes referred to as ergogenic aids. Cognitive performance-enhancing drugs, commonly called nootropics, are sometimes used by students to improve academic performance. Performance-enhancing substances are also used by military personnel to enhance combat performance.
=== Peptides work === Pert published over 250 scientific articles on peptides and their receptors and the role of these neuropeptides in the immune system. She held a number of patents for modified peptides in the treatment of psoriasis, Alzheimer's disease, chronic fatigue syndrome, stroke and head trauma. One of her modified peptides, Peptide T, had been considered for the treatment of AIDS and neuroAIDS. A placebo-controlled, three-site, 200+ patient NIH-funded clinical trial which was principally concerned with possible neurocognitive improvements, was conducted between 1990 and 1995. It was found that the effect of Peptide T was not significantly different from that of placebo on the primary end points of the study - various aspects of brain function. However, Peptide T was associated with improved performance (memory and learning) in the subgroup of patients with more severe cognitive impairment. A long-delayed analysis of antiviral effects from the NIH study showed peripheral viral load (combined plasma and serum) was significantly reduced in the DAPTA-treated group. An eleven-person study of Peptide T effects on cellular viral load showed reductions in infected monocyte reservoir to undetectable levels in most of the patients. Pert was developing orally active peptide anti-inflammatory treatments for pain and Alzheimer's Disease and studies for treatment of HIV persistent viral reservoirs.

After longer oxidation periods, sheets were permeable to salt but not larger organic molecules. In 2015, a team fabricated a graphene oxide home water purification system resembling a "tea bag" that, over the course of a day, can remove 95% of heavy metals when immersed in a drinking-water pot. A composite comprising small NiFe2O4 ferrimagnetic nanoparticles and partially reduced graphene oxide functionalized with nitrogen atoms was successfully used to remove Cr(III) ions from water. The advantage of this nanocomposite is that it can be magnetically separated from water. One project layered carbon atoms in a honeycomb structure, forming a hexagon-shaped crystal that measured about 0.1 millimeters in width and length, with subnanometer holes. Later work increased the membrane size to on the order of several millimeters. Graphene attached to a polycarbonate support structure was initially effective at removing salt. However, defects formed in the graphene. Filling larger defects with nylon and small defects with hafnium metal, followed by a layer of oxide, restored the filtration effect. In 2016, engineers developed graphene-based films powered by the sun that can filter dirty/salty water. Bacteria were used to produce a material consisting of two nanocellulose layers. The lower layer contains pristine cellulose, while the top layer contains cellulose and graphene oxide, which absorb sunlight and produce heat.

Actin, gamma-enteric smooth muscle is a protein that in humans is encoded by the ACTG2 gene. Actins are highly conserved proteins that are involved in various types of cell motility, and maintenance of the cytoskeleton. In vertebrates, three main groups of actin isoforms, alpha, beta and gamma have been identified. The alpha actins are found in muscle tissues and are a major constituent of the contractile apparatus. The beta and gamma actins co-exist in most cell types as components of the cytoskeleton, and as mediators of internal cell motility. Actin, gamma 2, encoded by this gene, is a smooth muscle actin found in enteric tissues. ACTG2 has been shown to interact with Emerin. Lawrencium is a synthetic chemical element; it has symbol Lr (formerly Lw) and atomic number 103. It is named after Ernest Lawrence, inventor of the cyclotron, a device used to discover many artificial radioactive elements. A radioactive metal, lawrencium is the eleventh transuranium element, the third transfermium, and the last member of the actinide series. Like all elements with atomic numbers over 100, lawrencium can only be produced in particle accelerators by bombarding lighter elements with charged particles. Fourteen isotopes of lawrencium are known; the most stable is 266Lr with half-life 11 hours, but the shorter-lived 260Lr (half-life 2.7 minutes) is most commonly used in chemistry because it can be produced on a larger scale. Chemistry experiments confirm that lawrencium behaves as a heavier homolog to lutetium in the periodic table, and is trivalent. It could thus also be classified as the first of the 7th-period transition metals. Its electron configuration is anomalous for its position in the periodic table, having an s2p configuration instead of the s2d configuration of its homolog lutetium. However, this does not appear to affect lawrencium's chemistry. In the 1950s, 1960s, and 1970s, many claims of synthesis of element 103 of varying quality were made by laboratories in the Soviet Union and the United States. The priority of the discovery and therefore the name of the element was disputed between Soviet and American scientists.
There are many physiological mechanisms that control starting and stopping a meal. The control of food intake is a physiologically complex, motivated behavioral system. Hormones such as cholecystokinin, bombesin, neurotensin, anorectin, calcitonin, enterostatin, leptin and corticotropin-releasing hormone have all been shown to suppress food intake. Eating rapidly leads to obesity and overeating, probably because the feelings of satiety can be slower.
==== Alliance for Germany ==== The Alliance for Germany presented its election manifesto under the slogan "Never again socialism" („Nie wieder Sozialismus“). Its main objectives included achieving German reunification based on the Basic Law for the Federal Republic of Germany as a unified constitution, guaranteeing rights to private property and unrestricted freedom of trade, and removing all barriers to Western investment. The alliance called for the immediate introduction of the Deutsche Mark at a 1:1 exchange rate to the East German mark. Social policy commitments included establishing a social security network, environmental protection measures, securing energy supplies, and harmonising laws with the West, notably abolishing criminal offences linked to political activities. Other priorities were monument preservation, education reform, maintaining day nurseries, restoring the federal states (Länder), and ensuring press freedom.
== Chemistry == The oxidation reaction that converts adrenaline into adrenochrome occurs both in vivo and in vitro. Silver oxide (Ag2O) was among the first reagents employed for this, but a variety of other oxidizing agents have been used successfully. In solution, adrenochrome is pink and further oxidation of the compound causes it to polymerize into brown or black melanin compounds.
Nicotinamide riboside (NR, SR647) is a pyridine-nucleoside and a form of vitamin B3. It functions as a precursor to nicotinamide adenine dinucleotide, or NAD, through a two-step and a three-step pathway.

==== Methadone ==== Methadone is a commonly used full-opioid agonist in the treatment of opioid use disorder. It is effective in relieving withdrawal symptoms and cravings in people with opioid addiction, and can also be used in pain control in certain situations. While methadone is a widely prescribed form of OAT, it often requires more frequent clinical visits compared to buprenorphine/naloxone, which also has a better safety profile and lower risk of respiratory depression and overdose. Important considerations when initiating methadone include the patient's opioid tolerance, the time since last opioid use, the type of opioid used (long-acting vs. short-acting), and the risk of methadone toxicity. Methadone comes in different forms: tablet, oral solution, or an injection. One of methadone's benefits is that it can last up to 56 hours in the body, so if a patient misses a daily dose, they will not typically struggle with withdrawal symptoms. Other advantages of methadone include reduction in infectious disease related to injection drug use, and reduced mortality. Methadone has a number of potential side effects, including slowed breathing, nausea, vomiting, restlessness, and headache.
Pholcodine is an opioid cough suppressant (antitussive). It helps suppress unproductive coughs and also has a mild sedative effect, but has little or no analgesic effects. It is also known as morpholinylethylmorphine and homocodeine. Pholcodine is found in certain cough lozenges, and more commonly as an oral solution, typically 5 mg / 5 ml. Adult dosage is 5-10 ml up to 3-4 times daily. Pholcodine now largely replaces the previously more common codeine linctus, as it has a much lower potential for dependence. Pholcodine has been widely used as an antitussive agent but by 2023 concerns over its association with anaphylaxis in some circumstances meant that it has been withdrawn from sale in many territories. Pholcodine is not prescribed in the United States where it is classed as a Schedule I drug, the most highly controlled drug category. Following the conclusion of a review of post-marketing safety data by the Medicines and Healthcare products Regulatory Agency, all pholcodine-containing medicines were recalled and withdrawn from the UK as a precaution. The available data has demonstrated that pholcodine use, particularly in the twelve months before general anesthesia with NMBAs (neuromuscular blocking agents), is a risk factor for developing an anaphylactic reaction to NMBAs. In December 2022, the European Medicines Agency recommended their withdrawal in the EU. As of February 2023, the Australian Therapeutic Goods Administration canceled the registration of pholcodine.
== Public health concerns == Epidemics of fatal pulmonary hypertension and heart valve damage associated with pharmaceutical anorectic agents have led to the withdrawal of products from the market. This was the case with aminorex in the 1960s, and again in the 1990s with fenfluramine (see: Fen-phen). Likewise, association of the related appetite suppressant phenylpropanolamine with hemorrhagic stroke led the Food and Drug Administration (FDA) to request its withdrawal from the market in the United States in 2000, and similar concerns regarding ephedrine resulted in an FDA ban on its inclusion in dietary supplements in 2004. A Federal judge later overturned this ban in 2005 during a challenge by supplement maker Nutraceuticals. It is also debatable as to whether the ephedrine ban had more to do with its use as a precursor in methamphetamine manufacture rather than health concerns with the ingredient as such.

The result is a much longer duration of action than the native hormone.
The fatty diacid chain associates strongly with serum albumin, which keeps the peptide in circulation and shields it from rapid renal clearance. Bound peptide is released gradually, producing a sustained receptor signal. This mechanism also reduces the peak-to-trough variation seen with shorter-acting analogues.
The balance between peripheral receptor activation and signalling in the central nervous system is not fully resolved. The extent to which slowed gastric emptying accounts for reduced energy intake, compared with direct effects on appetite circuits, is debated. Long-term effects on lean mass and on tissues outside the gut and brain are still under study.
