Semaglutide is a synthetic peptide that acts as an agonist at the glucagon-like peptide-1 receptor. It is a structural analogue of human GLP-1(7-37), modified to resist enzymatic degradation by dipeptidyl peptidase-4. The peptide backbone contains alpha-aminoisobutyric acid at position 8, a substitution that stabilises the helix and slows cleavage. A fatty diacid side chain attached through a linker at lysine 34 promotes binding to serum albumin, which extends the circulating half-life. These two modifications together allow less frequent administration than native GLP-1 requires.
Activation of the GLP-1 receptor couples to Gs signalling and raises intracellular cyclic AMP in pancreatic beta cells. The resulting insulin release depends on prevailing glucose concentrations, so the effect is greater when glucose is elevated. Receptor engagement also suppresses glucagon secretion and slows gastric emptying, which flattens post-meal glucose excursions. In the central nervous system, signalling in hypothalamic and brainstem regions is associated with reduced appetite and lower energy intake. Studies continue to examine effects on cardiac, renal and hepatic endpoints; whether those benefits are independent of weight change remains an open question.
Semaglutide is a synthetic peptide analog of glucagon-like peptide-1, a hormone released from intestinal L-cells after food intake. It contains 31 amino acids and differs from native GLP-1 through modifications that slow enzymatic breakdown. The peptide was developed to extend the short circulating half-life of endogenous GLP-1, which is measured in minutes. Researchers introduced the compound in the early 2010s. Two backbone changes and a fatty acid side chain define its structure, distinguishing it from earlier GLP-1 receptor agonists.
The compound binds the GLP-1 receptor on pancreatic beta cells and other tissues, activating a G-protein signaling cascade that raises intracellular cyclic AMP. This action increases glucose-dependent insulin secretion when blood glucose is elevated, while binding also slows gastric emptying and reduces glucagon release. In the central nervous system, receptor activation in the hypothalamus and brainstem contributes to reduced appetite. The fatty acid chain binds albumin, which protects the peptide from renal filtration and enzymatic degradation. This albumin binding is central to its extended circulation time.

Two principal therapeutic variants exist under separate regulatory filings, one indicated for glycemic control in type 2 diabetes and one for chronic weight management. Both use the same active molecule; differences lie in formulation strength, titration schedule, and labeling. Regulatory agencies in the United States and European Union approved injectable forms in 2017 and 2018 respectively. An oral tablet formulation received approval later, using a carrier molecule to enhance absorption across the gastric epithelium. Labeling differs by jurisdiction and by indication.
The distinction between established facts and open questions matters here. That the peptide binds the GLP-1 receptor and stimulates insulin release in a glucose-dependent manner is well documented. How individual variability in receptor density, gastric emptying rate, and gut microbiome composition shapes response remains an active research area. Long-term outcomes beyond five years of continuous use are not yet fully characterized in published trials, and several extension studies are ongoing.
Quantification and purity assessment rely on separation methods coupled to optical or mass detection. Reversed-phase high-performance liquid chromatography resolves the intact peptide from related impurities and is the standard assay technique. Size-exclusion chromatography measures aggregates, while ion-exchange chromatography separates charge variants produced by deamidation. Mass spectrometry confirms identity and detects mass shifts of a few daltons. In biological matrices, liquid chromatography with tandem mass spectrometry is often used because immunoassays can cross-react with endogenous GLP-1 or with circulating fragments.
As a peptide, semaglutide is sensitive to conditions that break amide bonds or modify side chains. Deamidation of asparagine and glutamine residues, oxidation of methionine and tryptophan, and non-covalent aggregation are the main degradation routes described in published stability work. Rate depends strongly on pH, buffer species, ionic strength, temperature and exposure to light. Formulators therefore choose a defined solution pH and often add excipients such as phosphate buffer, propylene glycol and phenol, each of which plays a separate role in pH control, tonicity or preservation.
Storage guidance for the finished injectable product distinguishes the unused state from the in-use state. Before first use, pens are kept refrigerated between 2 and 8 degrees Celsius, protected from light, and never frozen, since freezing can disrupt the peptide or the device. After first use, label instructions in several markets permit storage at room temperature up to about 30 degrees Celsius for a limited number of days. Solid research-grade material is normally held at or below minus 20 degrees Celsius, often with desiccant, and allowed to equilibrate before opening.
=== Addiction and dependence === Although classified as a central nervous system (CNS) stimulant, the addiction and dependence liabilities of modafinil are considered low. The exact mechanisms of action of modafinil are not known, and its pharmacological profile differs from that of the classical stimulants such as cocaine or amphetamine. Although modafinil shares biochemical mechanisms with stimulant drugs, it is less likely to have mood-elevating properties. The similarities in effects with caffeine are not clearly established. Unlike other stimulants, modafinil does not induce a strong subjective feeling of pleasure or reward or euphoria, which contributes to its lower abuse potential. Albeit to a lower degree than classical stimulants, modafinil still can produce psychoactive, euphoric, and subjective effects typical for abused stimulants. Despite initial assessments of low abuse potential, some evidence indicates modafinil acts on the same neurobiological mechanisms as other addictive stimulants, which is a reason for caution when prescribing. Modafinil scores lower than amphetamine on standardized abuse-potential assessments, suggesting reduced propensity for abuse. The US Drug Enforcement Administration has classified modafinil as a Schedule IV controlled substance; the medicine is recognized for having valid medical uses with low addiction potential. The International Narcotics Control Board does not classify it as a narcotic or a psychotropic substance.
Three structural isomers of L-tyrosine are known. In addition to the common amino acid L-tyrosine, which is the para isomer (para-tyr, p-tyr or 4-hydroxyphenylalanine), there are two additional regioisomers, namely meta-tyrosine (also known as 3-hydroxyphenylalanine, L-m-tyrosine, and m-tyr) and ortho-tyrosine (o-tyr or 2-hydroxyphenylalanine), that occur in nature. The m-tyr and o-tyr isomers, which are rare, arise through non-enzymatic free-radical hydroxylation of phenylalanine under conditions of oxidative stress. They are toxic to both animals and plants.
== Adhesive contact between elastic bodies == When two solid surfaces are brought into close proximity, they experience attractive van der Waals forces. R. S. Bradley's van der Waals model provides a means of calculating the tensile force between two rigid spheres with perfectly smooth surfaces. The Hertzian model of contact does not consider adhesion possible. However, in the late 1960s, several contradictions were observed when the Hertz theory was compared with experiments involving contact between rubber and glass spheres. It was observed that, though Hertz theory applied at large loads, at low loads

== Production == To make kimchi, cabbage or daikon is sliced into small, uniform pieces to increase surface area. The pieces are then coated with salt to draw out water and increase salt content, which helps preserve them by preventing the growth of harmful microorganisms. This salting process can use 5–7% salt for 12 hours or 15% salt for 3–7 hours. After salting, excess water is drained and the seasoning ingredients are added. Sugar can be used to bind any remaining water. Finally, the brined vegetables are packed into airtight jars and left to ferment at room temperature for 24 to 48 hours. The ideal salt concentration during fermentation is about 3%. Since the fermentation process results in the production of carbon dioxide, the jar should be "burped" daily to release the gas. The more fermentation that occurs, the more carbon dioxide will be incorporated, which results in a very carbonated-drink-like effect.
Physiology of muscle contraction involves several interactions. Myosin filaments act as molecular motors and by binding to actin enables filament sliding. Furthermore, members of the skeletal muscle lipid droplet-associated proteins family associate with other proteins, as activator of adipose triglyceride lipase and its coactivator comparative gene identification-58, to regulate lipolysis in skeletal muscle
MD simulation of the full satellite tobacco mosaic virus (STMV) (2006, Size: 1 million atoms, Simulation time: 50 ns, program: NAMD) This virus is a small, icosahedral plant virus that worsens the symptoms of infection by Tobacco Mosaic Virus (TMV). Molecular dynamics simulations were used to probe the mechanisms of viral assembly. The entire STMV particle consists of 60 identical copies of one protein that make up the viral capsid (coating), and a 1063 nucleotide single stranded RNA genome. One key finding is that the capsid is very unstable when there is no RNA inside. The simulation would take one 2006 desktop computer around 35 years to complete. It was thus done in many processors in parallel with continuous communication between them. Folding simulations of the Villin Headpiece in all-atom detail (2006, Size: 20,000 atoms; Simulation time: 500 μs= 500,000 ns, Program: Folding@home) This simulation was run in 200,000 CPU's of participating personal computers around the world. These computers had the Folding@home program installed, a large-scale distributed computing effort coordinated by Vijay Pande at Stanford University. The kinetic properties of the Villin Headpiece protein were probed by using many independent, short trajectories run by CPU's without continuous real-time communication. One method employed was the Pfold value analysis, which measures the probability of folding before unfolding of a specific starting conformation. Pfold gives information about transition state structures and an ordering of conformations along the folding pathway.
=== Advanced treatment plant technology === For some emerging contaminants, several advanced technologies—sonolysis, photocatalysis, Fenton-based oxidation and ozonation—have treated pollutants in laboratory experiments. Another technology is "enhanced coagulation" in which the treatment entity would work to optimize filtration by removing precursors to contamination through treatment. In the case of THMs, this meant lowering the pH, increasing the feed rate of coagulants, and encouraging domestic systems to operate with activated carbon filters and apparatuses that can perform reverse osmosis. Although these methods are effective, they are costly, and there have been many instances of treatment plants being resistant to pay for the removal of pollution, especially if it wasn't created in the water treatment process as many EC's occur from runoff, past pollution sources, and personal care products. It is also difficult to incentivize states to have their own policies surrounding contamination because it can be burdensome for states to pay for screening and prevention processes. There is also an element of environmental injustice, in that lower income communities with less purchasing and political power cannot buy their own system for filtration and are regularly exposed to harmful compounds in drinking water and food. However, recent treads for light-based systems shows great potential for such applications. With the decrease in cost of UV-LED systems and growing prevalence of solar powered systems, it shows great potential to remove CEC while keeping costs low.

The second step of the process, producing the actual creatine molecule, occurs solely in the cytosol, where the second enzyme, S-adenosylmethionine:guanidinoacetate methyltransferase (GAMT), is found. The creatine is then transported through the bloodstream and taken up through sodium-dependent creatine transporters by cells that require creatine.
=== Peptides work === Pert published over 250 scientific articles on peptides and their receptors and the role of these neuropeptides in the immune system. She held a number of patents for modified peptides in the treatment of psoriasis, Alzheimer's disease, chronic fatigue syndrome, stroke and head trauma. One of her modified peptides, Peptide T, had been considered for the treatment of AIDS and neuroAIDS. A placebo-controlled, three-site, 200+ patient NIH-funded clinical trial which was principally concerned with possible neurocognitive improvements, was conducted between 1990 and 1995. It was found that the effect of Peptide T was not significantly different from that of placebo on the primary end points of the study - various aspects of brain function. However, Peptide T was associated with improved performance (memory and learning) in the subgroup of patients with more severe cognitive impairment. A long-delayed analysis of antiviral effects from the NIH study showed peripheral viral load (combined plasma and serum) was significantly reduced in the DAPTA-treated group. An eleven-person study of Peptide T effects on cellular viral load showed reductions in infected monocyte reservoir to undetectable levels in most of the patients. Pert was developing orally active peptide anti-inflammatory treatments for pain and Alzheimer's Disease and studies for treatment of HIV persistent viral reservoirs.

=== Recreational use === Over-the-counter pseudoephedrine has been misused as a psychostimulant. Six case reports and one case series of pseudoephedrine misuse have been published as of 2021. There is a case report of self-medication with pseudoephedrine for treatment of depression.
== Reception == Half-Life 2: Deathmatch was well-received upon its release, with reviewers praising its inclusion of the gravity gun. GameSpot lauded the game for the Source engine's "impressive use of physics" and the available selection of maps. IGN paid similar compliments to the title, also speaking highly of the "fast play style that [Half-Life 2: Deathmatch] produces." Multiple reviews stated that the game suffered from large amount of lag upon release, but GameSpy noted that this was remedied soon after as more servers came online.

It belongs to the incretin mimetic class and acts as a long-acting glucagon-like peptide-1 receptor agonist. The class includes several peptides with different half-lives and routes of administration.
The side chain enables reversible binding to albumin in the bloodstream, which protects the peptide from rapid renal clearance. This extends the interval between administrations compared with unmodified GLP-1.
No. It shares much of the native sequence but carries substitutions and an added side chain. These changes increase stability against enzymatic breakdown.
It is a synthetic analog of GLP-1 produced through medicinal chemistry to resist enzymatic degradation. The design goal was longer circulation than the native hormone.
