As a peptide, semaglutide is sensitive to conditions that break amide bonds or modify side chains. Deamidation of asparagine and glutamine residues, oxidation of methionine and tryptophan, and non-covalent aggregation are the main degradation routes described in published stability work. Rate depends strongly on pH, buffer species, ionic strength, temperature and exposure to light. Formulators therefore choose a defined solution pH and often add excipients such as phosphate buffer, propylene glycol and phenol, each of which plays a separate role in pH control, tonicity or preservation.
Storage guidance for the finished injectable product distinguishes the unused state from the in-use state. Before first use, pens are kept refrigerated between 2 and 8 degrees Celsius, protected from light, and never frozen, since freezing can disrupt the peptide or the device. After first use, label instructions in several markets permit storage at room temperature up to about 30 degrees Celsius for a limited number of days. Solid research-grade material is normally held at or below minus 20 degrees Celsius, often with desiccant, and allowed to equilibrate before opening.
Quantification and purity assessment rely on separation methods coupled to optical or mass detection. Reversed-phase high-performance liquid chromatography resolves the intact peptide from related impurities and is the standard assay technique. Size-exclusion chromatography measures aggregates, while ion-exchange chromatography separates charge variants produced by deamidation. Mass spectrometry confirms identity and detects mass shifts of a few daltons. In biological matrices, liquid chromatography with tandem mass spectrometry is often used because immunoassays can cross-react with endogenous GLP-1 or with circulating fragments.
Reverse-phase high-performance liquid chromatography with ultraviolet detection near 214 nm is the standard purity method, reported as area percent. Mass spectrometry, usually with electrospray ionisation, confirms identity and reveals covalent modifications. Size-exclusion chromatography quantifies aggregates and fragments. Peptide mapping after enzymatic digestion localises changes to specific sequence regions. Circular dichroism and infrared spectroscopy report on secondary structure, while light scattering tracks particle formation in liquid formulations. No single technique captures every quality attribute.
Quality control relies on pharmacopoeial monographs where they exist, combined with in-house specifications for identity, purity, water content, and counter-ion composition. Reference standards allow calibration across laboratories, although certified materials for every analogue are not universally obtainable. Batch records, chromatograms, and mass spectra form the documentation trail. Regulatory classification varies by jurisdiction and intended use, and research-grade material differs from pharmaceutical-grade material in testing scope. Analytical uncertainty is often expressed as relative standard deviation across replicate injections.
Lyophilised semaglutide is generally held at -20 °C or below, protected from light and moisture. Reconstituted solutions are typically kept at 2-8 °C and used within a defined window because degradation accumulates over time. Repeated freeze-thaw cycles are discouraged, since each cycle can promote aggregation and reduce monomeric content. Room-temperature stability of the solid has been examined in some studies but remains incompletely characterised for long durations, so cold storage is the conservative default for research material.

Native GLP-1 is degraded rapidly by dipeptidyl peptidase-4. Semaglutide resists this cleavage because alanine at position 8 is replaced by alpha-aminoisobutyric acid. A second substitution at position 34 introduces arginine, which further stabilizes the peptide. The most distinctive modification is a spacer and C18 fatty diacid attached at lysine 26, enabling strong albumin affinity. These three changes together produce a half-life measured in days rather than minutes, and the same structural logic underlies other long-acting analogs in this class.
Semaglutide is a synthetic peptide analog of glucagon-like peptide-1, a hormone released from intestinal L-cells after food intake. It contains 31 amino acids and differs from native GLP-1 through modifications that slow enzymatic breakdown. The peptide was developed to extend the short circulating half-life of endogenous GLP-1, which is measured in minutes. Researchers introduced the compound in the early 2010s. Two backbone changes and a fatty acid side chain define its structure, distinguishing it from earlier GLP-1 receptor agonists.

== Roles in disease and medicine == It has been found to have anticarcinogenic properties. In a study on mice, injecting a preparation of DSIP over the mice's lifetime decreased total spontaneous tumor incidence 2.6-fold. The same study found it to also have geroprotective effects: it slowed down the age-related switching-off of oestrous function; it decreased by 22.6% the frequency of chromosome aberrations in bone marrow cells and it increased by 24.1% maximum life span in comparison with the control group. Levels of DSIP may be significant in patients diagnosed with major depressive disorder (MDD). In several studies, levels of DSIP in the plasma and cerebrospinal fluid are significantly deviated from the norm in patients with MDD, though there are contradictions as to whether levels are higher or lower than healthy control patients. Studies have demonstrated a direct link between GILZ expression (homologous to DSIP) and adipogenesis which has links to obesity and metabolic syndrome. In studies on rats with metaphit-induced epilepsy DSIP acted as an anticonvulsant, significantly decreasing the incidence and duration of fits suggesting DSIP as a potential treatment for epilepsy. DSIP has been found to have an analgesic effect. In studies on mice it was found to have a potent antinociceptive effect when administered intracerebroventricularly or intracisternally (see: Route of administration). Due to its possible effects on sleep and nociception, trials have been carried out to determine whether DSIP can be used as an anaesthetic.
She carried only a little money, but before she left, Hahn gave her a diamond ring he had inherited from his mother. Meitner continued to correspond with Hahn by mail. In late 1938 Hahn and Strassmann found evidence of isotopes of an alkaline earth metal in their sample. Finding a group 2 metal was problematic, because it did not logically fit with the other elements found thus far. Hahn initially suspected it to be radium, produced by splitting off two alpha-particles from the uranium nucleus, but chipping off two alpha particles via this process was unlikely. The idea of turning uranium into barium (by removing around 100 nucleons) was seen as preposterous. During a visit to Copenhagen on 10 November, Hahn discussed these results with Niels Bohr, Meitner, and Otto Robert Frisch. Further refinements of the technique, leading to the decisive experiment on 16–17 December 1938, produced puzzling results: the three isotopes consistently behaved not as radium, but as barium. Hahn, who did not inform the physicists in his Institute, described the results exclusively in a letter to Meitner on 19 December:
== Classification == As a definition of "cell type" is yet to be agreed, it is not possible yet to arrive at a precise number of human cell types. There is, for example, significant variation in these cell types depending on the specific surface proteins they possess. An extensive listing of human cell types was published by Vickaryous and Hall in 2006, collecting 411 different types of human cells, including 145 types of neurons. The Human Cell Atlas project, which started in 2016, had as one of its goals to "catalog all cell types (for example, immune cells or brain cells) and sub-types in the human body". By 2018, the Human Cell Atlas description based the project on the assumption that "our characterization of the hundreds of types and subtypes of cells in the human body is limited", but the word hundreds was removed in later versions. On 2021, Stephen Quake guessed that the upper limit of the number of human cell types would be around 6000, based on a reasoning that "if biologists had discovered only 5% of cell types in the human body, then the upper limit of cell types to discover is somewhere around 6000 (i.e., 300/0.05)." Other different efforts have used different numbers. A count of cells in the human body published in 2023 divided the cells in about 400 types to perform the calculation.

The compound was first administered to two normal human volunteers by Abass Alavi in August 1976 at the University of Pennsylvania. Brain images obtained with an ordinary (non-PET) nuclear scanner demonstrated the concentration of FDG in that organ. Later, the substance was used in dedicated positron tomographic scanners, to yield the modern procedure. The logical extension of positron instrumentation was a design using two two-dimensional arrays. PC-I was the first instrument using this concept and was designed in 1968, completed in 1969 and reported in 1972. The first applications of PC-I in tomographic mode as distinguished from the computed tomographic mode were reported in 1970. It soon became clear to many of those involved in PET development that a circular or cylindrical array of detectors was the logical next step in PET instrumentation. Although many investigators took this approach, James Robertson and Zang-Hee Cho were the first to propose a ring system that has become the prototype of the current shape of PET. The first multislice cylindrical array PET scanner was completed in 1974 at the Mallinckrodt Institute of Radiology by the group led by Ter-Pogossian. The PET–CT scanner, attributed to David Townsend and Ronald Nutt, was named by Time as the medical invention of the year in 2000.

== Chemistry == Three chemical syntheses of zolpidem are common. 4-Methylacetophenone is first brominated and that product is treated with 2-amino-5-methylpyridine to give the imidazopyridine. From here the reactions use a variety of reagents to complete the synthesis, either involving thionyl chloride or sodium cyanide. These reagents are challenging to handle and require thorough safety assessments. Though such safety procedures are common in the industry, they make clandestine manufacture difficult. Several major side-products of the sodium cyanide reaction have been characterised and include dimers and mannich products. Alpidem is also an imidazopyridine and is an analogue of zolpidem. Both agents are GABAA receptor positive allosteric modulators. However, whereas zolpidem is used as a hypnotic and sedative, alpidem was used as an anxiolytic.
23 October U.S. field commanders denied that there had been any changes in strategy and tactics, other than not making ground assaults on fortified PAVN/VC base areas which would be bombed instead. Many claimed to have been unaware of the concept of "protective reaction" and while the battlefield situation had reduced in intensity, it was nowhere near a ceasefire.
==== Expedition 70/71 ==== Caldwell Dyson returned to flight training as a backup crew member for the Soyuz MS-24. She backed up her NASA astronaut colleague Loral O'Hara. She was officially assigned to the Soyuz MS-25 in 2023. Her first attempt to launch on the Soyuz MS-25 mission on March 21, 2024 was aborted with 20 seconds left before lift-off. The scrub occurred because of a low voltage reading in the Soyuz rocket electrical system. She launched on March 23, 2024 with Roscosmos cosmonaut Oleg Novitsky and Belarusian cosmonaut Marina Vasilevskaya. After a two-day rendezvous profile, the Soyuz crew docked with International Space Station's Prichal module on March 25, 2024. The first weeks of her mission dealt with the SpaceX CRS-30 mission, as well as viewing the total eclipse above North America on April 8, 2024 and the redocking of SpaceX Crew-8 in early May. Starting in mid-May, Expedition 71 crew focused themselves on the preparation for the three planned EVAs. During this period, the crew checked out EMUs, tools, and reviewed procedures. They also performed maintenance tasks around station as well as continued scientific work. On June 6, Caldwell Dyson saw the arrival of the first crewed mission of Starliner. The Starliner crew was composed of Barry Wilmore and Sunita Williams. Caldwell Dyson monitored the arrival of Starliner alongside Matthew Dominick and helped the Starliner crew to accomplish their test flight objectives including testing the spacecraft habitability for four crew members and its capability as a safe haven in case of an emergency.

Gorgonzola blue cheese takes its name from the village of Gorgonzola in Italy where it was first made. Belonging to the family of Stracchino cheeses, Gorgonzola is a whole milk, white, and "uncooked" cheese. This blue cheese is inoculated with Penicillium glaucum which, during ripening, produces the characteristic of blue-green veins. There are two variants with different odor: natural and creamy Gorgonzola. 63 components in natural Gorgonzola cheese and 52 components in creamy Gorgonzola cheese contribute to odor with 2-nonanone, 1-octen-3-ol, 2-heptanol, ethyl hexanoate, methylanisole and 2-heptanone being the prominent compounds for odor in both cheeses.
Several such models for mineral-mediated polymerization have emerged, such as the interlayers of layered double hydroxides like green rust over wet-dry cycles. Some scenarios for peptide formation have been proposed that are even compatible with aqueous solutions, such as the hydrophobic air-water interface and a novel "sulfide-mediated α-aminonitrile ligation" scheme, where amino acid precursors come together to form peptides. Polymerization of life's building blocks is an active area of research in prebiotic chemistry.
Eurycoma longifolia (commonly called tongkat ali, Malaysian ginseng or long jack) is a flowering plant in the family Simaroubaceae. It is native to Indochina (Cambodia, Laos, Malaysia, Myanmar, Thailand and Vietnam) and Indonesia (the islands of Borneo and Sumatra), but has also been found in the Philippines. The plant is a medium-sized slender shrub that can reach 10 m (33 ft) in height, and is often unbranched. The root has been used in traditional medicine in Southeast Asia over centuries. It is also a widely used dietary supplement by bodybuilders, who believe that it increases testosterone levels and athletic performance, although there is no clinical evidence for its effectiveness on health or any disease. Rare cases of liver injury have occurred from its use, mostly in bodybuilders.
=== Recreational drug use === Subcutaneous injection may also be used by people to (self-) administer recreational drugs. This can be referred to as skin popping. In some cases, the administration of illicit drugs in this way is associated with unsafe practices leading to infections and other adverse effects. In rare cases, this results in serious side effects such as AA amyloidosis. Recreational drugs reported to be administered subcutaneously have included cocaine, mephedrone, and amphetamine derivatives such as PMMA.

Ice formation concentrates solutes and can mechanically stress the peptide or damage the delivery device. Thawing afterwards may leave aggregates that are not visible to the eye. Refrigeration keeps the solution above its freezing point while slowing chemical degradation.
Size-exclusion chromatography is the most common technique, separating monomer from dimer and higher-order species by hydrodynamic volume. It is usually paired with ultraviolet detection and, for trace work, with multi-angle light scattering. Results depend on mobile-phase conditions, so method details matter when comparing datasets.
The detailed degradation profile under real-world temperature excursions and repeated handling is not fully described in the open literature. Relative contributions of photo-degradation versus thermal pathways are also incompletely mapped. Whether specific impurity species carry different biological activity is an open question rather than an established finding.
Reverse-phase high-performance liquid chromatography with ultraviolet detection is the usual choice, with results reported as area percent. Complementary methods such as size-exclusion chromatography and mass spectrometry are needed because a single separation cannot resolve every impurity class. Purity figures are therefore method dependent and should always be read alongside the technique used.
